Because of their actions at the opiate receptor, these opiate agonists and antagonists are also useful as diagnostic tools, and may also be useful for the treatment of other medical conditions such as endocrine, gastrointestinal, and central nervous disorders.
Opiate agonists act by binding to specific receptor sites in the brain and other tissues, and may have side effects due to the generalized activation of all endogenous opiate receptors. The opiate antagonist, in contrast, blocks the effect of the agonist at the receptor. Thus, opiate antagonists are of value in controlling undesirable physiological actions of opiate agonists and in the treatment of opiate addiction and for use as analgesics.
There is a need for selective and specific antagonists of mu (.mu.) opiate receptors in the nervous system, where the.mu. receptor population may be involved in the regulation of physiological responses associated with pain and gastrointestinal secretion. The advantage of such antagonists is that they would not produce the undesirable side effects produced by non-selective opiate antagonists such as respiratory depression, constipation and sedation.
A wide variety of 3,4-dihydro-4-oxo-1H-isoquinolines have been reported as having opioid antagonist activity. For example, U.S. Pat. No. 3,916,895 to Safer and Sessler (1975) describes 3,4-dihydro-4-oxo-1H-isoquinolines that are reported to be useful as analgesics.
A variety of 3,4-dihydro-4-oxo-1H-isoquinolines, useful as.mu.-opiate receptor antagonists, are disclosed in U.S. Pat. No. 4,447,445 to Robinson et al (1984).
